ADF and cofilin-1 collaborate to promote cortical actin flow and the leader bleb-based migration of confined cells

چکیده

Melanoma cells have been shown to undergo fast amoeboid (leader bleb-based) migration, requiring a single large bleb for migration. In leader blebs, is rapid flow of cortical actin that drives the cell forward. Using RNAi, we find co-depleting cofilin-1 and depolymerizing factor (ADF) led increase in actin, suggesting both proteins regulate actin. Furthermore, severing factors can promote contractility through regulation architecture. However, RNAi but not ADF significant decrease stiffness. We found be enriched at necks, whereas reduced sizes frequency motile cells. Strikingly, without had blebs with abnormally long necks. Many these failed retract displayed slow turnover. Collectively, our data identifies as remodeling required

The ADF/Cofilin-Pathway and Actin Dynamics in Podocyte Injury

ADF/cofilins are the major regulators of actin dynamics in mammalian cells. The activation of ADF/cofilins is controlled by a variety of regulatory mechanisms. Dysregulation of ADF/cofilin may result in loss of a precisely organized actin cytoskeletal architecture and can reduce podocyte migration and motility. Recent studies suggest that cofilin-1 can be regulated through several extracellular...

Erk regulation of actin capping and bundling by Eps 8 promotes cortex tension and leader bleb - 1 based migration

15 Within the confines of tissues, cancer cells can use blebs to migrate. Eps8 is an actin bundling and 16 capping protein whose capping activity is inhibited by Erk, a key MAP kinase that is activated by 17 oncogenic signaling. We tested the hypothesis that Eps8 acts as an Erk effector to modulate actin 18 cortex mechanics and thereby mediate bleb-based migration of cancer cells. Cells confine...

Roles of ADF/cofilin in actin polymerization and beyond

In collaboration or competition with many other actin-binding proteins, the actin-depolymerizing factor/cofilins integrate transmembrane signals to coordinate the spatial and temporal organization of actin filament assembly/disassembly (dynamics). In addition, newly discovered effects of these proteins in lipid metabolism, gene regulation, and apoptosis suggest that their roles go well beyond r...

Actin Depolymerizing Factor (ADF/Cofilin) Enhances the Rate of Filament Turnover: Implication in Actin-based Motility

Actin-binding proteins of the actin depolymerizing factor (ADF)/cofilin family are thought to control actin-based motile processes. ADF1 from Arabidopsis thaliana appears to be a good model that is functionally similar to other members of the family. The function of ADF in actin dynamics has been examined using a combination of physical-chemical methods and actin-based motility assays, under ph...

The ADF/cofilin proteins: stimulus-responsive modulators of actin dynamics.

Whether a cell is harnessing actin polymerization to produce movement, preparing for cytokinesis, or assembling long-lived actin filament-based structures such as sarcomeres or microvilli, actin polymerization must be controlled in time and space. Changes in actin organization often occur on a time scale that dictates an intimate coupling of a signal transduction apparatus to the machinery that...
